The Birth Of GLP-1
The observation that the insulinotropic actions of GLP-1 are reduced in type 2 diabetes mellitus (T2DM) led to the development of incretin-based therapies - GLP-1 receptor agonists and dipeptidyl peptidase 4 (DPP-4) inhibitors - for the treatment of hyperglycaemia in these patients. Here, we review the role of GLP-1 and the actions of associated therapies on glucose metabolism, the gut-renal axis, classical renal risk factors, and renal end points in randomized controlled trials of GLP-1 receptor agonists and DPP-4 inhibitors in patients with T2DM. These actions collectively contribute to its role in glucose regulation and hormone-based weight loss appetite control, highlighting its importance in metabolic treatments. Eating protein can help your body release amino acids and peptides, which are both important to GLP-1 secretion.10 Protein is also essential to appetite regulation and better glycemic control in those with type 2 diabetes or at risk for the condition. The gastrointestinal tract - the largest endocrine network in human physiology - orchestrates signals from the external environment to maintain neural and hormonal control of homeostasis. Adult male Sprague-Dawley rats (Charles River Laboratories, USA) were individually housed in double-wide cages with a temperature and humidity-controlled environment on a 12-hour/12-hour light-dark cycle.
Counting was accomplished at 20X magnification and numerical aperture of 0.75, using a light/fluorescent microscope (Nikon Eclipse Ni, Nikon, USA) and Imaris software version 8.1.2 (Bitplane, Switzerland). The statistical analyses were performed using Prism 5.0 software (GraphPad, San Diego, CA, USA). Then, sections were rinsed in PBS and mounted onto Superfrost Plus glass slides (Thermo Scientific, Waltham, appetite regulation MA USA) and coverslipped with DAPI mounting medium. Tissues were washed in PBS and incubated for 2 h with fluorescent secondary antibody Alexa Fluor 594 donkey anti-rabbit (1:500, Jackson ImmunoResearch, USA). Briefly, sections were washed in 50% EtOH and subsequently incubated for 20 min in 1% sodium borohydride. F3020) in RPMI media for 45 min at 37 °C. After incubation, media was collected, centrifuged at 1000 rpm for 5 min at 4 °C and supernatant stored at −80 °C until further analysis. Plasma was stored at −80 °C until ELISA assay. Brains were cut into 30-μm coronal sections on a cryostat (Leica microsystem) and stored in cryoprotectant until processing for immunohistochemistry. Brains were immediately removed and flash-frozen in cold isopentane to preserve RNA. Brains were collected and maintained overnight in 4% PFA.
In this study, we performed real-time PCR with the islets of db/db diabetic mice at different ages, and found that the expression levels of Glp1r were comparable to those of the islets of nondiabetic db/misty controls at the age of four weeks, and were significantly decreased at the age of eight and 12 weeks. Before you start taking GLP-1 agonist medicines, it is important that your diabetic eye screening is up to date. Considerable interest exists in identifying effects of these drugs beyond glucose-lowering, possibly resulting in improved macrovascular and microvascular outcomes, including in diabetic kidney disease.
